Springbok scrumhalf Cobus Reinach will earn his 50th Test cap when he takes the field against Wales at the Principality Stadium in Cardiff on Saturday.
Coach Rassie Erasmus named an experienced 23-player team with 525 caps, in spite of not having all his initial traveling squad available for selection.
A number of players had to return to their clubs and were not available for selection.
The starting team features eight changes from the team that defeated Ireland 24-13 in Dublin last week.
Three forwards and four backs were retained.
Erasmus opted for a seven-one split in favour of the forwards on the bench.
